Overview

Galiximab is a primatized (cynomolgus monkey variable region, human constant region) IgG1 monoclonal antibody that targets CD80, a costimulatory molecule expressed on the surface of B cells and certain lymphoma cells. By binding to CD80, galiximab blocks its interaction with CD28 on T-cells, thereby modulating immune responses. Its primary mechanism involves inducing antibody-dependent cell-mediated cytotoxicity (ADCC) against malignant B cells. Galiximab has been investigated mainly for follicular non-Hodgkin lymphoma and other B-cell lymphomas, as well as autoimmune disorders such as psoriasis and relapsed Hodgkin lymphoma. The drug was developed by Biogen Idec (now Biogen). Clinical trials showed modest single-agent activity in follicular lymphoma but higher response rates when combined with rituximab or chemotherapy. Development for all indications has been discontinued[1][2][3][4][5][7].
