Gallium-68 DOTA-HK-011 is a radiopharmaceutical diagnostic agent designed for positron emission tomography (PET) imaging. It consists of the positron-emitting isotope gallium-68 chelated by DOTA and conjugated to HK-011, a small-molecule ligand that specifically targets fibroblast activation protein (FAP). FAP is highly expressed on the surface of cancer-associated fibroblasts (CAFs) in the stroma of various epithelial tumors but has low expression in normal tissues. By binding to FAP, this tracer allows for the visualization and quantification of FAP expression, which is useful for tumor staging, monitoring treatment response, and identifying candidates for FAP-targeted radioligand therapy (theranostics). HK-011 is a quinoline-based derivative developed to optimize the pharmacokinetic profile and imaging contrast compared to earlier FAP-targeted tracers.
