Gallium-68 DOTA-SIGLEC-9 is an investigational radiopharmaceutical PET tracer developed by Turku University Hospital for the non-invasive imaging of inflammation. The agent comprises a peptide sequence derived from the Sialic acid-binding immunoglobulin-type lectin 9 (Siglec-9) protein, which is a natural ligand for Vascular Adhesion Protein-1 (VAP-1), also known as Amine oxidase copper containing 3 (AOC3). VAP-1 is an endothelial cell surface enzyme that mediates leukocyte recruitment and is significantly upregulated in inflamed tissues but absent from healthy vasculature. By chelating the positron-emitting isotope gallium-68 via a DOTA linker to the Siglec-9 peptide, the tracer enables high-contrast PET imaging of active inflammatory sites. It is currently being evaluated in clinical trials for the assessment of rheumatoid arthritis, various forms of vasculitis, and pulmonary sarcoidosis to aid in diagnosis and monitoring of disease activity.
