Gallium Ga 68 NOTA-DNA multivalent SGC8 is an experimental radiopharmaceutical designed for positron emission tomography (PET) imaging of malignancies that overexpress protein tyrosine kinase 7 (PTK7). The agent consists of the positron-emitting radioisotope Gallium-68 ($^{68}$Ga) chelated by NOTA (1,4,7-triazacyclononane-1,4,7-triacetic acid) and conjugated to a multivalent DNA scaffold containing multiple copies of the SGC8 aptamer. By utilizing a multivalent approach—often through DNA nanostructures like tetrahedrons or branched DNA—the tracer achieves significantly higher binding affinity and improved tumor retention compared to monovalent aptamer sequences. The SGC8 aptamer specifically targets the extracellular domain of PTK7, a cell surface pseudokinase frequently upregulated in T-cell acute lymphoblastic leukemia (T-ALL) and various solid tumors, making it a valuable biomarker for cancer diagnosis and staging.
