Drug intelligence / Profile preview
GalNAc-siCNNM4 (also referred to as GalNAc-siCnnm4) is a liver-targeted small interfering RNA (siRNA) therapeutic conjugated to N-acetylgalactosamine (GalNAc). It is designed to selectively silence the expression of Cyclin M4 (CNNM4), a magnesium exporter, in hepatocytes. Developed in collaboration with Silence Therapeutics using their proprietary mRNAi GOLD platform, GalNAc-siCNNM4 aims to restore intracellular magnesium homeostasis, alleviate endoplasmic reticulum stress, and reduce inflammation and fibrosis. It is currently in preclinical development for various liver conditions, including metabolic dysfunction-associated steatohepatitis (MASH/NASH), alcohol-associated liver disease (ALD), primary biliary cholangitis (PBC), primary sclerosing cholangitis (PSC), and acetaminophen-induced liver injury.
