Drug intelligence / Profile preview
GalNAc-siJag1 is a hepatocyte-targeted small interfering RNA (siRNA) modified with N-acetylgalactosamine (GalNAc) that targets Jagged1 (JAG1). Developed by Sanofi in collaboration with Columbia University, GalNAc-siJag1 is designed to selectively knock down JAG1 expression in hepatocytes, thereby inhibiting the aberrant hepatocyte-to-hepatic stellate cell (HSC) Notch signaling pathway. This targeted gene silencing aims to reduce liver fibrosis and inflammation associated with metabolic dysfunction-associated steatohepatitis (MASH), formerly known as nonalcoholic steatohepatitis (NASH). Preclinical studies in mouse models of MASH/NASH have demonstrated that subcutaneous administration of GalNAc-siJag1 significantly decreases liver JAG1 expression, reduces Notch target gene activation, and ameliorates or reverses established liver fibrosis.
