GALNAC4S-6ST siRNA-1 is a small interfering RNA (siRNA) designed to silence the expression of the *CHST15* gene, which encodes the enzyme N-acetylgalactosamine 4-sulfate 6-O-sulfotransferase (GALNAC4S-6ST). This enzyme is a critical regulator of glycosaminoglycan sulfation, specifically responsible for the biosynthesis of the chondroitin sulfate E (CS-E) motif by adding a sulfate group to the C6 position of GalNAc(4S). In the context of castration-resistant prostate cancer (CRPC), CS-E has been identified as a driver of androgen-independent growth by activating the IL-6/STAT3 signaling pathway. By utilizing the RNA interference (RNAi) pathway to knock down GALNAC4S-6ST, this siRNA reduces CS-E levels, thereby inhibiting downstream oncogenic signaling and potentially limiting tumor progression in advanced prostate cancer. It is primarily utilized in preclinical research to investigate the therapeutic potential of targeting specific sulfation patterns in the tumor microenvironment.
