GALNAC4S-6ST siRNA-2 is an experimental small interfering RNA (siRNA) designed to silence the expression of the *CHST15* gene, which encodes the enzyme N-acetylgalactosamine 4-sulfate 6-O-sulfotransferase (GALNAC4S-6ST). This enzyme is responsible for the 6-O-sulfation of chondroitin 4-sulfate to produce chondroitin sulfate E (CS-E). In the context of castration-resistant prostate cancer (CRPC), GALNAC4S-6ST is upregulated and drives the production of CS-E, which in turn activates IL-6/STAT3 signaling to promote androgen-independent tumor growth. By knocking down GALNAC4S-6ST, this siRNA reduces CS-E levels and inhibits oncogenic signaling pathways, potentially offering a therapeutic strategy for CRPC.
