Gamgertamig is an investigational humanized IgG4 bispecific T cell–engager antibody targeting the B-cell maturation antigen (BCMA, TNFRSF17) and the CD3 epsilon chain on T cells, designed to deplete pathogenic BCMA-expressing B cells and plasma cells via T cell–dependent cellular cytotoxicity in oncology and immune-mediated diseases.[1][4][5] It incorporates a “detuned” CD3-binding arm engineered to reduce T cell cytokine release while preserving cytotoxic activity, aiming to expand the therapeutic index and mitigate severe immune activation.[1][2] Initially developed by Keymed Biomedical Technology (Chengdu) and now being advanced by Ouro Medicines, gamgertamig has been studied in multiple myeloma and related plasma cell dyscrasias, and is currently in a global Phase 1b basket trial in autoimmune cytopenias; it has received U.S. FDA Orphan Drug Designation for autoimmune hemolytic anemia and immune thrombocytopenia.[1][2][5]
