Overview

Gamma-hydroxybutyrate (GHB) is a naturally occurring neurotransmitter and a small molecule central nervous system depressant being investigated by the Psychiatric University Hospital, Zurich, for the prevention of post-traumatic stress disorder (PTSD). In the TRAUMA-PRO clinical trial, GHB is administered as a post-exposure prophylaxis to healthy volunteers following an experimental trauma film to enhance slow-wave sleep (deep sleep). The therapeutic rationale is that boosting deep sleep shortly after a traumatic event facilitates the healthy consolidation of memories, thereby reducing the frequency and intensity of subsequent intrusive memories, which are a hallmark of PTSD. GHB acts primarily as a high-affinity agonist at the GHB receptor and a weak agonist at the gamma-aminobutyric acid type B (GABA-B) receptor.
