Overview

Gamma-irradiated BCG vaccine is an experimental, inactivated version of the live attenuated Bacillus Calmette-Guérin (BCG) vaccine, specifically utilizing the Danish strain 1331 from Statens Serum Institut (SSI). Developed and investigated by Radboud University Medical Center, this formulation is designed to induce "trained immunity"—a process of epigenetic and metabolic reprogramming of innate immune cells such as monocytes and natural killer cells. This reprogramming results in an enhanced, non-specific immune response to various heterologous infections. The use of gamma irradiation serves to inactivate the bacterium, potentially making it safer for use in immunocompromised populations who cannot receive the live attenuated vaccine, while preserving the structural and molecular components necessary to train the innate immune system.
