Gamma-PN is an investigational universal vaccine developed for the prevention of Streptococcal pneumonia, caused by *Streptococcus pneumoniae*. It is a whole-cell, inactivated, gamma-irradiated, and unencapsulated strain (Rx1) of *S. pneumoniae*, genetically modified by deletion of the autolysin gene (lytA) and replacement of the pneumolysin gene (ply) with a non-toxic pneumolysoid allele (PdT). The vaccine is administered via intranasal or intramuscular routes. Unlike current vaccines that cover up to 23 serotypes, Gamma-PN aims to provide broad protection against all known pneumococcal types as well as future emerging strains. Its mechanism involves stimulating both innate and adaptive immune responses without adjuvant; it induces dose-dependent rises in serum IgG levels and activates neutrophils consistent with immunostimulation[1][3][6][8]. Developed by GPN Vaccines in collaboration with the University of Adelaide, Gamma-PN is currently in Phase II clinical trials for Streptococcal pneumonia.
