gamma-polyglutamic acid-cisplatin complex is a polymer-drug conjugate formulation designed for the controlled release of the chemotherapeutic agent cisplatin. Developed by researchers at Osaka University, the formulation utilizes gamma-polyglutamic acid (gamma-PGA), a biodegradable and non-immunogenic polymer, to form a complex with cisplatin. This delivery system aims to provide a sustained release of the drug into the bloodstream, thereby reducing the peak plasma concentrations that contribute to acute renal toxicity (nephrotoxicity) while maintaining or enhancing anti-tumor efficacy. Preclinical studies in murine mesothelioma models have demonstrated that the complex maintains therapeutic blood levels longer than free cisplatin and shows superior anti-tumor activity with reduced adverse effects.
