gamma-tocopherol-rich tocopherol mixture (γ-TmT) is a natural product mixture composed of various forms of vitamin E, typically containing approximately 57% gamma-tocopherol, 24% delta-tocopherol, 13% alpha-tocopherol, and 1.5% beta-tocopherol. Derived as a by-product of vegetable oil refining, γ-TmT has been investigated preclinically and clinically for its anti-inflammatory, anti-oxidative, and anti-carcinogenic properties. It has demonstrated efficacy in animal models of colon, lung, mammary, and prostate cancers, as well as colon inflammation, and has been evaluated in early-phase clinical trials for asthma, airway inflammation, and prostate cancer. Its mechanism of action involves the inhibition of cyclooxygenase-1 (COX-1), cyclooxygenase-2 (COX-2), and 5-lipoxygenase (5-LOX), leading to a reduction in inflammatory mediators such as prostaglandin E2 (PGE2) and leukotriene B4 (LTB4). Additionally, γ-TmT traps reactive nitrogen species, reduces oxidative stress, and modulates downstream signaling pathways (including Nrf2, PTEN, AKT, and Caspase-3) to induce apoptosis and inhibit cell proliferation in cancer cells.
