Ganetespib + docetaxel is a combination therapy investigated primarily for advanced non-small cell lung cancer (NSCLC), particularly adenocarcinoma. Ganetespib is a small molecule inhibitor of heat shock protein 90 (Hsp90), a molecular chaperone involved in the stabilization and function of multiple oncogenic proteins. By inhibiting Hsp90, ganetespib disrupts several signaling pathways critical for cancer cell survival and proliferation[2][4][5]. Docetaxel is a taxane-class chemotherapeutic agent that promotes microtubule assembly and prevents their depolymerization, thereby inhibiting mitosis and inducing apoptosis in rapidly dividing cells[1][6]. The combination was evaluated in phase III clinical trials as second-line therapy for patients with advanced NSCLC after platinum-based chemotherapy. However, the addition of ganetespib to docetaxel did not improve overall survival compared to docetaxel alone, leading to early termination of pivotal studies due to futility[2][8].
