Overview

**GANIT** is an investigational intramuscular plasmid-DNA gene-therapy system designed for insulin-deficient diabetes. It delivers a construct to skeletal muscle that encodes **INSA-F**, an insulin aspart analogue engineered with furin-cleavage sites. A glucose-responsive promoter containing nuclear factor of activated T-cells regulatory motifs controls INSA-F transcription, aiming to couple insulin analogue production to extracellular glucose concentrations. In a type 1 diabetes mouse study, two treatments over two months improved glucose homeostasis, glucose tolerance, and glycated haemoglobin and reduced oxidative stress, cardiac fibrosis, and renal fibrosis. GANIT remains a preclinical research-stage therapeutic system.
