Overview

Ganitumab is a fully human monoclonal antibody that targets the type 1 insulin-like growth factor receptor (IGF1R). By binding to IGF1R, ganitumab blocks the interaction of IGF1 and IGF2 with their receptor, thereby inhibiting downstream signaling pathways such as PI3K/Akt that are important for tumor cell proliferation and survival. This mechanism leads to inhibition of tumor growth and induction of cancer cell death. Ganitumab was originally developed by Amgen and has been investigated in multiple cancers, including Ewing sarcoma, breast cancer, colorectal cancer, pancreatic cancer, rhabdomyosarcoma, non-small cell lung cancer, small cell lung cancer, prostate cancer, and other solid tumors. It received orphan drug designation for Ewing sarcoma but has not demonstrated significant clinical benefit in phase III trials for this or other indications[2][3][6][7][9].
