Drug intelligence / Profile preview
Ganoderic acid DM is a triterpenoid compound isolated from the fruiting body of the medicinal mushroom *Ganoderma lucidum* (Lingzhi). It is a bioactive natural product classified as a lanostane-type triterpenoid. Ganoderic acid DM possesses **antiandrogenic, anti-osteoclastogenic, anti-cancer, and pro-apoptotic activity**. Mechanistically, it inhibits key mediators including c-Fos and NFATc1, consequently suppressing dendritic cell-specific transmembrane protein (DC-STAMP) and reducing osteoclast fusion. It also acts as an inhibitor of androgen receptor signaling and has demonstrated toxicity against both androgen-dependent and -independent prostate cancer cells, making it a potential agent for advanced prostate cancer. Recent studies highlight its **potential in breast cancer by inhibiting PIK3CA and related pathways, suggesting cell cycle arrest, apoptosis induction, and reduced cancer cell growth**. Additional inhibitory effects on 5α-reductase, PARP, CDK, NF-κB, and TNF-α have been reported, indicating broad implications for cancer therapy and immunomodulation[1][3][5][7][9].
