Drug intelligence / Profile preview
**GAP19 + GAP26** is a combination of two synthetic connexin 43 (Cx43) mimetic peptides, Gap19 and Gap26, which are widely used as research reagents to selectively inhibit Cx43 hemichannels. Gap19 is a nonapeptide (KQIEIKKFK) derived from the cytoplasmic loop of Cx43 that prevents the interaction between the C-terminus and the cytoplasmic loop, a prerequisite for hemichannel gating, without affecting gap junctional communication. Gap26 is a 13-amino acid peptide (VCYDKSFPISHVR) homologous to the first extracellular loop of Cx43 that blocks both hemichannels and, with longer exposure, gap junctions. Together or individually, these peptides are investigated in preclinical models of myocardial infarction, cerebral ischemia, spinal cord injury, and renal fibrosis (such as unilateral ureteral obstruction) to prevent pathological hemichannel opening, ATP release, and downstream inflammatory processes like macrophage pyroptosis.
