Overview

Gardasil 9 is a 9-valent recombinant human papillomavirus (HPV) vaccine developed by Merck & Co. It is composed of purified virus-like particles (VLPs) of the major capsid (L1) protein of nine HPV types: 6, 11, 16, 18, 31, 33, 45, 52, and 58. The vaccine works by inducing a humoral immune response, producing neutralizing antibodies that prevent the virus from entering host cells. It is widely approved for the prevention of cervical, vulvar, vaginal, and anal cancers, as well as genital warts and oropharyngeal cancer. In a specific Phase III clinical trial (NCT04255849) led by Weill Medical College of Cornell University, the vaccine is being evaluated for its efficacy in preventing persistent oral HPV infection in adult men living with HIV, which serves as a surrogate endpoint for the prevention of oropharyngeal and other head and neck cancers.
