Drug intelligence / Profile preview
GAS6-OVA (GO) is an engineered recombinant fusion protein consisting of the efferocytic bridge protein growth arrest-specific 6 (GAS6) fused to the model antigen ovalbumin (OVA). Developed by researchers at the University of Chicago and New York University, GAS6-OVA is utilized as a preclinical research tool to investigate the mechanisms of efferocytosis-mediated antigen-specific immune tolerance. The fusion protein binds to phosphatidylserine (PS) on apoptotic cells and targets TAM receptors (such as Axl, Mer, and Tyro3) on antigen-presenting cells (APCs), enhancing dendritic cell uptake and antigen presentation. In vivo, GAS6-OVA drives bidirectional tolerogenic programming between dendritic cells and T cells, promoting regulatory T cell (Treg) enrichment and terminal exhaustion of CD8+ T cells, which prevents allergic airway pathology in mouse models of asthma.
