Overview

Gasdermin D short hairpin RNA (shGSDMD) is a research-grade RNA interference (RNAi) tool designed to specifically silence the expression of the gasdermin D (GSDMD) gene. GSDMD is a critical executioner of pyroptosis, a pro-inflammatory form of programmed cell death. Upon activation by inflammatory caspases, the N-terminal fragment of GSDMD oligomerizes to form large pores in the plasma membrane, leading to cell swelling, lysis, and the release of cytosolic contents such as IL-1β and LDH. In preclinical research, particularly in models of endometrial carcinoma, shGSDMD is utilized to investigate the role of pyroptosis in tumor biology and to evaluate the impact of GSDMD depletion on membrane integrity and inflammatory signaling. By reducing GSDMD levels, researchers can demonstrate the inhibition of pore formation and the subsequent reduction in pyroptotic cell death markers.
