A **gastrin siRNA polyplex** is a therapeutic nanoparticle formulation that complexes small interfering RNA (siRNA) targeting the **gastrin gene** with a cationic polymer to form a polyplex (micellar nanoparticle). This drug is designed to selectively deliver gastrin-targeting siRNA to tumors that overexpress the **cholecystokinin B (CCK-B) receptor**, notably in pancreatic ductal adenocarcinoma (PDAC). Upon cellular uptake mediated by CCK-B receptor targeting, the siRNA is released in tumor cells, resulting in **gene knockdown of gastrin**, leading to reduced peptide expression, inhibition of tumor growth, and block of metastasis in preclinical PDAC models. This approach minimizes off-target toxicity and achieves efficient, tumor-selective gene silencing[1].
