Overview

GATC-1021 is an AI-identified small molecule being developed for the treatment of opioid use disorder (OUD). Discovered through GATC Health's Operon platform in collaboration with the University of California, Irvine, the compound acts as a dual modulator of the 5-HT2A and 5-HT6 receptors. Preclinical data published in PNAS indicate that GATC-1021 reduces fentanyl self-administration and enhances neuroplasticity in the prefrontal cortex, as evidenced by increased thin dendritic spine density and Bdnf expression. Unlike many serotonergic agents, GATC-1021 demonstrates a non-hallucinogenic behavioral profile, offering a potential non-opioid therapeutic pathway that avoids the clinical and regulatory complexities of psychedelic-assisted therapies.
