Gatipotuzumab is a humanized monoclonal antibody that targets the tumor-specific epitope of mucin-1 (TA-MUC1), enabling it to differentiate between tumor MUC1 and non-tumor MUC1 epitopes. It works by binding to TA-MUC1 expressed on the cell surface of tumor cells, activating the immune system to induce antibody-dependent cellular cytotoxicity (ADCC) and phagocytosis against TA-MUC1-expressing tumor cells. The antibody is glyco-engineered using glyco-optimized production cell lines on the GlycoExpress platform, which provides a "human-like" glycosylation that increases immunogenicity and triggers a stronger immune response. Gatipotuzumab also becomes internalized into cancer cells by endocytosis once bound to TA-MUC1, allowing it to potentially be used as a drug conjugate to deliver toxins or radionuclides into cancer cells. It is being developed by Glycotope and NEMOD Biotherapeutics and has been investigated primarily for ovarian cancer.
