Gatipotuzumab + cetuximab is a **combination of two monoclonal antibodies** investigated in advanced refractory solid tumors, particularly in metastatic colorectal cancer, non-small-cell lung cancer, head and neck cancer, and breast cancer. **Gatipotuzumab** is a humanized, glyco-engineered antibody targeting the tumor-associated epitope of Mucin 1 (TA-MUC1), promoting antibody-dependent cellular cytotoxicity (ADCC) against TA-MUC1-expressing cancer cells. **Cetuximab** is a chimeric monoclonal antibody that targets the epidermal growth factor receptor (EGFR), inhibiting ligand binding and receptor activation, leading to inhibition of tumor cell growth, survival, and proliferation. The combination aims to jointly target TA-MUC1 and EGFR, both implicated in tumor progression and immune evasion, and has shown a promising safety profile and preliminary antitumor activity in heavily pretreated, EGFR-positive metastatic cancers[1][3][5].
