Gatipotuzumab + tomuzotuximab is a combination of two humanized monoclonal antibodies developed for the treatment of advanced solid tumors. Gatipotuzumab targets the tumor-associated epitope of mucin-1 (TA-MUC1), a glycoepitope present on many carcinomas, enabling tumor-specific binding and immune activation. Tomuzotuximab is an immune-enhanced, glycooptimized monoclonal antibody against the epidermal growth factor receptor (EGFR), designed to improve antibody-dependent cellular cytotoxicity (ADCC) and reduce allergic reactions compared to cetuximab. The combination aims to exploit potential synergistic effects by dual targeting TA-MUC1 and EGFR, both implicated in epithelial tumor progression, with enhanced ADCC activity observed preclinically. Both antibodies were developed by Glycotope[5][6][8].
