Gatralimab is a next-generation humanized IgG1 monoclonal antibody that targets CD52 (also known as CAMPATH-1 antigen), a glycoprotein expressed on the surface of most human lymphocytes. It was developed to provide similar efficacy to alemtuzumab with potentially reduced infusion/injection-associated reactions and decreased immunogenicity. Gatralimab induces lymphocyte depletion through antibody-dependent cell-mediated cytotoxicity and complement-dependent cytotoxicity. In clinical trials for progressive multiple sclerosis (MS), it produced dose-dependent depletion of lymphocytes and CD4+ regulatory T cells with partial repopulation over time. Although it demonstrated an acceptable safety profile and desired pharmacodynamic effects in phase 1 studies, development for MS was discontinued due to business reasons unrelated to safety or efficacy[2][3][7].
