Overview

This is a combination therapy comprised of **gavocabtagene autoleucel** (TC-210, gavo-cel), **nivolumab**, and **ipilimumab**. - **Gavocabtagene autoleucel** is an autologous T-cell therapy in which patient T cells are genetically engineered to express a single-domain anti-mesothelin antibody fused to the CD3ε subunit (T cell receptor fusion construct, TRuC), enabling the entire endogenous TCR complex to recognize and kill mesothelin-expressing tumor cells in an HLA-unrestricted manner[1][2][4]. - **Nivolumab** is a human IgG4 monoclonal antibody that blocks PD-1 (Programmed death-1) on T cells, preventing tumor-induced T-cell inhibition. - **Ipilimumab** is a human monoclonal antibody that blocks CTLA-4 (Cytotoxic T-lymphocyte-associated antigen 4), also enhancing T-cell activation. The primary indication, as tested in ongoing trials, is for **mesothelin-expressing solid tumors** such as malignant pleural mesothelioma, ovarian cancer, cholangiocarcinoma, and non-small cell lung cancer. The rationale for combining these agents is that the checkpoint inhibitors (nivolumab and ipilimumab) may enhance the anti-tumor activity and persistence of the engineered T cells by overcoming resistance mechanisms in the tumor microenvironment[1][2].
