GB-0669 is a novel, half-life extended monoclonal antibody developed using generative AI by Generate Biomedicines. It targets the highly conserved S2 stem helix region of the spike protein of SARS-CoV-2 and related sarbecoviruses. The antibody was designed to provide broad neutralization against current and future variants of SARS-CoV-2, with a focus on prophylactic use in populations at high risk for severe COVID-19 outcomes, such as immunocompromised individuals. Preclinical and phase 1 clinical data demonstrate potent viral neutralization across all major tested variants, dose-proportional pharmacokinetics with an estimated half-life between 55–70 days, and a favorable safety profile with only mild adverse events observed. A single intravenous dose has been projected to offer up to six months of protection against infection. Development as a prophylactic has been paused due to market conditions; further development for treatment or other indications is under consideration[1][2][4][5][6][7][8].
