Overview

GB-102 is a sustained-release, intravitreal depot formulation of the small molecule tyrosine kinase inhibitor sunitinib malate. It is designed for controlled, long-term delivery to the eye via microparticles that degrade over approximately six months. The drug acts as a pan-inhibitor of vascular endothelial growth factor (VEGF) receptors 1, 2, and 3, as well as other receptor tyrosine kinases including platelet-derived growth factor receptors (PDGFR) alpha and beta, colony-stimulating factor receptor (CSF1R), Fms-like tyrosine kinase 3 (FLT3), proto-oncogene protein c-kit (KIT), and proto-oncogene protein c-ret (RET). By inhibiting these targets, GB-102 blocks multiple angiogenic pathways implicated in retinal diseases such as wet age-related macular degeneration (wet AMD), diabetic macular edema, retinal vein occlusion, and diabetic retinopathy. Developed originally by Graybug Vision and now by CalciMedica following a merger in March 2023, GB-102 aims to reduce treatment burden by extending dosing intervals compared to current anti–VEGF therapies[1][4][7][8].
