Overview

GB-401 is a first-in-class, biodegradable intravitreal implant formulation containing a novel prodrug of timolol, a beta-adrenergic antagonist. It is designed for sustained release and long-term reduction of intraocular pressure (IOP) in patients with primary open-angle glaucoma (POAG). The implant is administered via injection into the eye using a proprietary applicator and aims to provide therapeutic effects for up to six months per dose. The prodrug utilizes hydrophobic linkers that degrade by hydrolysis, releasing active timolol over time. Preclinical studies have demonstrated sustained ocular drug levels, significant IOP reduction in animal models, and good ocular safety profiles. The goal of this approach is to improve patient compliance compared to daily topical therapies by reducing dosing frequency[1][3][4][5][6].
