GB-4362 is a systemically administered monoclonal antibody developed by Generate Biomedicines using its AI-powered generative protein design platform. It is designed to selectively bind and neutralize free monomethyl auristatin E (MMAE), a potent cytotoxic payload frequently used in antibody-drug conjugates (ADCs). By sequestering free MMAE released into the systemic circulation, GB-4362 aims to reduce payload-mediated toxicities, such as peripheral neuropathy and neutropenia, thereby improving the therapeutic index of MMAE-based ADCs. It is intended as an adjunctive therapy to be used in combination with ADCs like enfortumab vedotin to preserve anti-tumor activity while mitigating systemic side effects.
