GB-5267 is an investigational, armored chimeric antigen receptor (CAR) T-cell therapy targeting Mucin-16 (MUC16), a glycoprotein frequently overexpressed in ovarian cancer. Developed by Generate Biomedicines using its proprietary generative AI platform in collaboration with Roswell Park Comprehensive Cancer Center, the therapy is specifically engineered to address the challenges of the solid tumor microenvironment. It features a high-affinity MUC16 binder and is "armored" with interleukin-18 (IL-18) to enhance T-cell activation, proliferation, and persistence. The therapy is initially being developed for platinum-resistant metastatic ovarian cancer. The FDA cleared the Investigational New Drug (IND) application in December 2025, with Phase 1 clinical trials anticipated to begin in 2026.
