Overview

GB00408 is a novel, small molecule modulator of the Aryl Hydrocarbon Receptor (AhR) being developed for the treatment of autoimmune and inflammatory skin diseases, such as psoriasis and atopic dermatitis. Developed by Galderma in collaboration with Sirenas, GB00408 targets the AhR, a ligand-activated transcription factor that serves as a central regulator of immune responses and skin barrier function. By modulating AhR activity, the drug aims to restore immune homeostasis by influencing the differentiation and function of regulatory and effector T cells. Preclinical studies have demonstrated that GB00408 dose-dependently inhibits the secretion of pro-inflammatory cytokines, including CCL2 and IL-8, in human keratinocytes stimulated by Th2, Th22, and Th17/TNF-alpha pathways. In vivo, topical administration of GB00408 has shown efficacy in reducing epidermal inflammation and IL-17 gene expression in murine models of psoriasis.
