Overview

GB00499 is a novel small molecule modulator of the Aryl Hydrocarbon Receptor (AhR) being developed by Gossamer Bio for the treatment of autoimmune and allergic diseases, with a primary focus on inflammatory skin conditions such as psoriasis and atopic dermatitis. The Aryl Hydrocarbon Receptor is a ligand-activated transcription factor that serves as a central regulator of immune responses and skin barrier function. By modulating AhR activity, GB00499 aims to restore immune balance and inhibit the production of pro-inflammatory cytokines. In preclinical studies presented at the AAD 2024 meeting, GB00499 demonstrated dose-dependent inhibition of Th2/Th22-induced CCL2 secretion and IL-17/TNF-alpha-induced IL-8 secretion in human keratinocytes, as well as significant reduction of inflammatory markers in human epidermis models of psoriasis.
