GB18-06 is a GDF15-targeting VHH nanobody developed for the treatment of cancer cachexia. It functions by binding to Growth Differentiation Factor 15 (GDF15) with high affinity, thereby inhibiting the GDF15-GFRAL signaling pathway and reducing downstream ERK and AKT phosphorylation levels. Preclinical studies have demonstrated its ability to alleviate weight loss and restore physical function in cancer and chemotherapy-induced cachexia models. GB18-06 is currently undergoing a first-in-human, randomized, double-blind, placebo-controlled, dose-escalation Phase 1 clinical trial to evaluate its safety, tolerability, pharmacokinetics, pharmacodynamics, and immunogenicity in healthy participants, with a focus on its effect on circulating GDF15 concentrations and body weight.
