GB201 (also referred to as GBD201) is a bispecific antibody developed by Genor Biopharma for the treatment of solid tumors. It is engineered as a symmetric, tetravalent molecule comprising a CCR8 monoclonal antibody and a CTLA-4 VHH nanobody. The drug's mechanism of action focuses on the selective depletion of CCR8+CTLA-4+ regulatory T cells (Tregs) within the tumor microenvironment, which are highly immunosuppressive, while sparing peripheral Tregs and conventional T cells. The CTLA-4 arm is designed as a partial blocker to minimize the systemic immune-related adverse events typically associated with full CTLA-4 inhibitors like ipilimumab. Preclinical data presented at SITC 2023 demonstrated potent anti-tumor efficacy and a favorable safety profile in humanized mouse models of bladder and colorectal cancer.
