GB262-saporin is an antibody-drug conjugate (ADC) consisting of the bispecific antibody GB262 conjugated to the toxin saporin. GB262 is designed to target Programmed Death-Ligand 1 (PD-L1) and CD55 (Complement Decay-Accelerating Factor). The antibody features a weaker binding arm for CD55 to minimize toxicity to healthy cells while maintaining internalization capability in cancer cells. Upon binding, CD55 facilitates the co-internalization of PD-L1 and the delivery of the saporin payload. Saporin is a type I ribosome-inactivating protein (RIP) that induces cell death by halting protein synthesis. This therapeutic candidate is being developed for the treatment of various cancers, including pancreatic cancer, by leveraging both immune checkpoint inhibition and direct cytotoxic delivery.
