GB263T is a novel trispecific antibody designed to target the epidermal growth factor receptor (EGFR) and two distinct epitopes of the hepatocyte growth factor receptor (cMET). The molecule incorporates two humanized VHH antibodies that sequentially bind to different cMET epitopes, enhancing tumor specificity and widening the safety window. This design allows for improved antigen-antibody endocytosis and more complete blockade of signal transduction pathways compared to benchmarks. Additionally, its Fc region is engineered for enhanced antibody-dependent cell-mediated cytotoxicity (ADCC). Preclinical studies have demonstrated robust anti-tumor activity in models with various EGFR/cMET alterations. Clinically, it is being evaluated in phase 1/2 trials for patients with advanced non-small cell lung cancer (NSCLC) who have progressed after prior EGFR TKI and platinum-based chemotherapy.
