GB264 is an investigational antibody-drug conjugate (ADC) designed to target Claudin 18.2 (CLDN18.2), a tight junction protein that is highly and selectively expressed in various solid tumors, including gastric, gastroesophageal junction, and pancreatic cancers. Developed by Antibody Therapeutics (a subsidiary of Genor Biopharma), GB264 consists of a monoclonal antibody specific to CLDN18.2 conjugated to a cytotoxic payload. The mechanism of action involves the selective binding of the antibody to CLDN18.2 on the surface of tumor cells, followed by internalization of the ADC and subsequent release of the payload, which induces cell death. As of early 2024, the program is in the IND-enabling stage, with development focused on the treatment of advanced solid tumors.
