Overview

GB3226 is a novel, potent, and selective small molecule inhibitor with dual activity against ENL-YEATS (Eleven-Nineteen Leukemia protein YEATS domain) and FLT3 (Fms-like tyrosine kinase 3). It is being developed for the treatment of acute myeloid leukemia (AML), particularly targeting multiple genetic subsets of AML, including those associated with resistance and relapse. Preclinical studies have demonstrated that GB3226 exhibits superior efficacy compared to both FLT3 inhibitors and menin inhibitors in animal models. Its dual mechanism allows for rapid therapeutic effects via FLT3 inhibition and sustained response through ENL-YEATS inhibition. The drug also shows potential for additive or synergistic activity when combined with standard-of-care treatments. Galecto acquired global rights to this program from Bridge Medicines in 2024–2025 and is advancing it toward an IND submission expected in Q1 2026[1][2][9].
