GBD209 is a trispecific antibody developed by Genor Biopharma that simultaneously targets three key immune checkpoints: Programmed Cell Death Protein 1 (PD-1), Cytotoxic T-Lymphocyte Associated Protein 4 (CTLA-4), and T-cell Immunoreceptor with Ig and ITIM domains (TIGIT). By synergistically blocking these pathways, GBD209 is designed to enhance anti-tumor T-cell activity more effectively than monotherapies or bispecific combinations. A distinctive feature of GBD209 is its engineered partial blockade of CTLA-4 interactions with its ligands CD80/CD86, which is designed to be highly dependent on PD-1 expression. This mechanism aims to localize CTLA-4 inhibition to the tumor microenvironment, thereby reducing peripheral toxicity and improving the safety profile compared to traditional CTLA-4 inhibitors like ipilimumab. Preclinical studies have demonstrated superior efficacy in animal models and significantly better tolerance in in vivo toxicity models.
