Overview

GBD218 is an investigational trispecific T-cell engager developed by Genor Biopharma for the treatment of multiple myeloma. It is designed to simultaneously target CD3 on T cells and two distinct antigens on myeloma cells: B-cell maturation antigen (BCMA) and G protein-coupled receptor class C group 5 member D (GPRC5D). This dual-targeting approach aims to overcome the heterogeneous expression of these markers in myeloma patients and prevent antigen-escape-mediated relapse. The antibody is constructed using a "1+1+1" format with "knob into hole" technology and a silenced IgG1 Fc region. To mitigate the risk of cytokine release syndrome (CRS), GBD218 utilizes a low-affinity anti-CD3 arm while maintaining high-potency binding to BCMA and GPRC5D.
