GBP-088 is an oral, gut-restricted small molecule inhibitor of the solute carrier family 26 member 3 (SLC26A3) transporter, also known as Down-Regulated in Adenoma (DRA). Developed by GondolaBio, the drug is intended for the treatment of hyperoxaluria and the prevention of calcium oxalate kidney stone disease. SLC26A3 is a key transporter in the intestinal epithelium that facilitates the absorption of dietary oxalate. By inhibiting this protein, GBP-088 reduces the amount of oxalate entering the bloodstream and subsequently the kidneys, thereby lowering urinary oxalate levels—a major risk factor for stone formation. The compound is designed to remain within the gastrointestinal tract to minimize systemic exposure and potential off-target effects. It is currently undergoing Phase 1 clinical evaluation to determine its safety, tolerability, and pharmacokinetic profile in healthy adults.
