GC-072 is a potent, orally bioavailable **4-oxoquinolizine antibiotic** developed as a therapeutic candidate for the treatment of **melioidosis** and serious infections caused by select biothreat pathogens, including **Burkholderia pseudomallei**, **Burkholderia mallei**, **Bacillus anthracis**, **Yersinia pestis**, and **Francisella tularensis**[1][3][5][6][9][11]. Its mechanism of action involves **selective inhibition of bacterial topoisomerases II (DNA gyrase)** and **IV**, with high potency against both wild-type and quinolone-resistant bacterial strains, especially B. pseudomallei[1][2][3]. GC-072 exhibits bactericidal activity both extracellularly and intracellularly, effectively killing bacteria within hours and showing reduced rates of resistance development compared to standard-of-care antibiotics like ceftazidime[2][3]. GC-072 is distinctive for oral administration and has shown robust preclinical efficacy in in vitro and murine models, promoting dose-dependent survival in lethal inhalational challenges with B. pseudomallei, and is less impacted by resistance mechanisms found in contemporary antibiotics. GC-072 was originally developed by Evolva and is now under Emergent BioSolutions following acquisition[5][7][9].
