Overview

GC027 is an "off-the-shelf" allogeneic chimeric antigen receptor T-cell (CAR-T) therapy targeting the CD7 antigen. It is designed for the treatment of T cell malignancies such as relapsed or refractory T-cell acute lymphoblastic leukemia (T-ALL) and precursor T-cell lymphoblastic leukaemia-lymphoma. Unlike autologous CAR-T therapies that require patient-derived cells, GC027 uses healthy donor-derived T cells engineered to express a universal anti-CD7 CAR construct. This approach enables rapid availability and circumvents challenges like fratricide and product contamination associated with targeting antigens expressed on normal T cells. In clinical studies, single infusions of GC027 have demonstrated robust in vivo expansion and high rates of minimal residual disease-negative complete remission in heavily pretreated patients with manageable toxicity profiles. The therapy was originally developed by Gracell Biotechnologies using their proprietary TruUCAR platform[1][3][5][7].
