Overview

GC182 is a humanized monoclonal antibody that targets **Glypican-3 (GPC3)**, a cell-surface oncofetal antigen that is highly overexpressed in **hepatocellular carcinoma (HCC)** and other solid tumors, while remaining largely absent in normal adult tissues. Developed by **Chugai Pharmaceutical** (a subsidiary of the Roche Group), GC182 was engineered to bind with high affinity to the C-terminal region of the GPC3 protein. Its primary mechanism of action is the induction of **antibody-dependent cellular cytotoxicity (ADCC)**, which recruits natural killer (NK) cells and other immune effectors to destroy GPC3-expressing malignant cells. While Chugai's other anti-GPC3 antibody, **codrituzumab (GC33)**, was the primary candidate advanced into Phase 2 clinical trials as a monotherapy, the GC182 clone has served as a critical foundation for next-generation therapeutic modalities. Specifically, the antigen-binding fragments of GC182 are utilized in the construction of **ERY974**, a bispecific T-cell engaging (BiTE) antibody targeting GPC3 and CD3, and have been incorporated into various **CAR-T cell therapy** research programs.
