GC197 is an investigational universal (allogeneic/off-the-shelf) chimeric antigen receptor T-cell (CAR-T) therapy developed by Gracell Biotechnologies (which was acquired by AstraZeneca in 2024). It is designed to target CD19-positive B-cell malignancies, such as B-cell acute lymphoblastic leukemia (B-ALL) and B-lymphoblastic lymphoma. GC197 is engineered using Gracell's proprietary TruUCAR platform, which utilizes CRISPR/Cas9 to disrupt CD7 and the T-cell receptor (TCR) to prevent graft-versus-host disease (GvHD) and CAR-T cell fratricide, while expressing a CAR targeting CD19 and a CAR targeting CD7 to suppress host-versus-graft (HvG) rejection. It has been evaluated in a Phase I/II clinical trial in China for patients with relapsed or refractory CD19-positive B-cell malignancies.
