Overview

GC4711 + GC4419 is a combination of two **selective superoxide dismutase mimetics**, both developed by Galera Therapeutics for use primarily in conjunction with **stereotactic body radiation therapy (SBRT)** for advanced cancers, notably **locally advanced or borderline resectable pancreatic cancer** and **non-small cell lung cancer**. These agents rapidly convert superoxide to hydrogen peroxide and oxygen, aiming to **improve the therapeutic efficacy of radiation therapy** by reducing radiation-induced toxicity in normal tissues and enhancing antitumor effects. GC4419, also known as avasopasem manganese, has been primarily investigated for the reduction of radiation-induced severe oral mucositis and esophagitis, while both have shown synergy with SBRT in preclinical and clinical studies, indicating **potential improvements in overall survival, progression-free survival, and surgical resection rates** when used in combination with SBRT[1][2][3][4].
